The FGL (Fibroblast Growth Factor-Like) ELISA Kit is a specialized assay designed for the quantitative measurement of FGL protein levels in various biological samples such as serum, plasma, cell culture supernatants, and tissue homogenates. FGL, also known as FGF-like protein, plays a significant role in cell proliferation, differentiation, and development, particularly in the context of nervous system function and repair, as well as in metabolic regulation.

Content of the Kit

  • Pre-coated 96-Well Microplate:
    • Wells coated with a specific monoclonal or polyclonal antibody against FGL, allowing for the capture of the target protein from the samples.
  • Standards:
    • Known concentrations of recombinant or purified FGL protein, used to generate a standard curve for quantifying FGL in unknown samples.
  • Sample Diluent:
    • A buffer solution used to dilute biological samples to optimal concentrations for accurate measurement.
  • Detection Antibody:
    • An antibody specific to a different epitope of the FGL protein, typically conjugated to biotin or directly to an enzyme like HRP.
  • Streptavidin-HRP Conjugate (if using a biotinylated detection antibody):
    • Binds to the biotinylated detection antibody, enabling detection through enzymatic color change.
  • Substrate Solution (TMB):
    • Tetramethylbenzidine, which reacts with HRP to produce a measurable color change.

Assay Procedure Summary

  • Sample and Standard Addition: Add standards and samples to the wells. FGL in the samples binds to the antibody coated on the microplate.
  • Wash: Unbound materials are removed by washing the wells.
  • Detection Antibody Addition: Add the detection antibody specific to FGL, which binds to the captured protein.
  • Wash: Unbound detection antibody is washed away.
  • HRP-Streptavidin Conjugate Addition: (If using a biotinylated detection antibody) Add HRP-streptavidin conjugate, which binds to the biotinylated detection antibody.
  • Wash: Remove unbound HRP-streptavidin conjugate by washing.

Performance Characteristics

  • Sensitivity: Capable of detecting FGL at low picogram per milliliter (pg/mL) levels.
  • Precision: The assay typically shows low intra-assay and inter-assay variability, with coefficients of variation (CV) below 10%.
  • Linearity: Exhibits a linear response over the standard curve range, ensuring accurate quantification across different sample concentrations.
  • Recovery: High recovery rates, typically above 80%, ensuring the assay's robustness across various sample types.
