The MYBL ELISA Kit is designed for the quantitative measurement of MYBL (Myb-like protein) levels in various biological samples, including serum, plasma, cell lysates, and tissue homogenates. MYBL proteins, which are part of the Myb family of transcription factors, play crucial roles in cell cycle regulation, differentiation, and apoptosis, making them significant in cancer research and developmental biology.
Content of the Kit
- Pre-coated 96-Well Microplate:
- Wells coated with a monoclonal or polyclonal antibody specific to MYBL, ensuring the capture of the target protein from samples.
- Standards:
- Prepared solutions with known concentrations of recombinant MYBL protein, used to generate a standard curve for quantification.
- Sample Diluent:
- Buffer used to dilute biological samples to appropriate concentrations for accurate measurement.
- Detection Antibody:
- An antibody specific to a different epitope of the MYBL protein, often conjugated to biotin or directly to an enzyme like HRP.
- Streptavidin-HRP Conjugate (if biotinylated detection antibody is used):
- Binds to the biotinylated detection antibody, allowing for subsequent colorimetric detection.
- Substrate Solution (TMB):
- Tetramethylbenzidine, which reacts with HRP to produce a measurable color change.
Assay Procedure Summary
- Sample and Standard Addition: Samples and standards are added to the microplate wells, where MYBL protein binds to the immobilized capture antibody.
- Wash: Unbound components are removed by washing the wells.
- Detection Antibody Addition: The detection antibody, which recognizes a different region of the MYBL protein, is added and binds to the captured MYBL.
- Wash: Unbound detection antibody is washed away.
- HRP-Streptavidin Conjugate Addition: (If a biotinylated detection antibody is used) HRP-conjugated streptavidin is added, binding to the biotin on the detection antibody.
- Wash: Excess HRP-streptavidin conjugate is removed through washing.
Performance Characteristics
- Sensitivity: Detects MYBL in the low picogram per milliliter (pg/mL) range.
- Precision: Intra-assay and inter-assay variability are typically low, with coefficients of variation (CV) under 10%.
- Linearity: Exhibits a linear response over the concentration range of the standard curve.
- Recovery: Typically exceeds 80%, indicating the assay's ability to accurately measure MYBL levels in various sample types.
Storage and Stability
- Storage: Store at 2-8°C for most components. Reconstituted standards and antibodies may require storage at -20°C for extended periods.
- Shelf Life: Typically 6-12 months under proper storage conditions.
